Cytoplasmic and plasma-gelsolins are supplied as natively

prepared, pure proteins. Gelsolins are readily soluble proteins,

requiring Ca++ for activation. Gelsolins bind to the barbed ends of

actin filaments, preventing exchange of monomers (capping).

 

Supplied in a stable form as lyophilized proteins, fully functional

after reconstitution.

Plasma gelsolin has a molecular mass of 83kDa and consists of six homologous

subdomains (S1-S6). Gelsolin is a Ca++-dependent lipid binding actin regulatory

protein, possessing three different actin modulating activities: severing of F-actin,

nucleation of actin polymerization and capping of filaments barbed end.

Gelsolin has three actin binding sites, two high affinity Ca++ binding sites and two

binding sites for PIP2.

= 50nM (barbed end).

Fragmentation activity: The fragmentation activity of gelsolin was determined

by Ostwald viscometry at 25°C. The specific viscosity of F-actin

(1mg/ml) is reduced by ~65% after addition of gelsolin at a molar ratio of 1:100.

The gelsolin stock solution was used as described below containing

1% disaccharides. F-actin containing 100mM KCl, 10mM Imidazole pH 7.4,

1mM ATP was mixed with 0.2mM CaCl2 (final concentration) prior to the addition

of gelsolin.

TL40 was obtained by thermolysin cleavage of smooth muscle gelsolin.

It consists of the N-terminal homologous subdomains S1-S3 (aa 1-361,

molecular mass: ~40kD) and is a Ca++- independent proteolytic fragment of native

gelsolin. TL40 is preferentially used in F-actin fragmentation assays in the presence

of EGTA.

by <45% after addition of TL40 at a molar ratio of 1:100 (TL40:A) within less than

2min. For this purpose the TL40 stock solution was used as described below

containing 1% disaccharides. F-actin (100mM KCl, 10mM Imidazole pH 7.4,

1mM ATP) was mixed with TL40 and instantly measured within a time course of

0-30min by ostwald viscometry.
