Fluorescent Actin-Conjugates
Rhodamine-actin conjugates are tools for investigating cytoskeleton dynamics. This fluorescent conjugates are prepared by labelling lysine residues of actin with activated esters of rhodamine dyes. Rhodamine actins have been widely used in fluorescence microscopical studies. Due to the superior features of modern fluorescence dyes characterized e.g. by significantly higher photostability, conjugates like Atto-actins are by far more suitable for these applications.

Rhodamine-Actin (rabbit skeletal muscle alpha actin)
Description Actin from rabbit skeletal muscle is a single chain polypeptide with a molecular weight of 42kD consisting of 375 amino acids. Rhodamine-actin is chemically modified G-actin by conugation of Cys374 with tetramethylrhodamine. Rhodamine-Actin (λem 554nm, λem 590nm) is >99% pure according to scanning densitometry from Coomassie Blue G-250 stained PAA-Gels and possesses the polymerization properties equal to native G-actin. The dye/protein molar ratio was determined to be 0.5:1.
Rhodamine-Actin is supplied as a lyophilized powder containing 2mM Tris-Cl pH 8.2, 0.4mM ATP and 0.1mM DTT, 1mM NaN3, 0.2mM CaCl2 and 0.3% disaccharides when reconstituted with H2O to a 1mg/ml (23.8µM) solution.
Product Storage and Stability The product stored at -70°C upon arrival and will be stable in performance for at least 6 months from the date of purchase. The solubilized product is kept on ice, protect from light and should be used within 1 week. Avoid refreezing of actin.
