Novel potent and selective mTOR inhibitor. Inhibits mTOR kinase with IC50 = 0.45 nM and PI3 kinase (PI3K), with p110α IC50 = 661 nM. It shows improved cellular potency LNCap IC50 = 42 nM as compared to WYE-354 (Cat # CD0270). WYE-23 microsomal stability T1/2 was shown to be > 30 min.

|
A cell-permeable pyrazolopyrimidine compound that acts as a potent and ATP competitive inhibitor of mTOR (IC50 = 4 nM). Active in LNCap cellular assay (IC50 = 355 nM). Selective against PI3K’s (p110a IC50 = 1026 nM, p110b, p110g, p110d > 6000 nM). Selective against other PIKK’s and other kinases. When injected into tumor-bearing nude mice, WYE-354 inhibited mTORC1 and mTORC2 and displayed robust antitumor activity in PTEN-null tumors.
