Latanoprost is a potent and selective agonist of Prostaglandin F receptor (FP). It activates FP, a G-protein
coupled receptor (GPCR), with EC50 of 10 nM, and with at least 500-fold selectivty over other Prostaglandin
receptors. Upon activation, FP promotes the decrease of intraocular pressure and contraction of smooth
muscle.
Clinically, Latanoprost (aka Xalatan(r)) is used as a topical ophthalmic solution for controlling the
progression of glaucoma or ocular hypertension by increasing the outflow of aqueous fluid from the eyes,
therefore reducing intraocular pressure.

LX7101 is a pyrrolopyrimidine-based, topically-delivered inhibitor of LIM domain kinase 2 (LIMK2), a kinase
associated with the regulation of intraocular pressure. LX7101 acts by enhancing fluid outflow through the
trabecular meshwork of the eye. Preclinical studies indicate that LX7101 significantly reduced intraocular
pressure in both mouse and monkey models. LX7101 recently completed a randomized, double-blind,
placebo-controlled Phase 1/2a trial for patients with open angle glaucoma or ocular hypertension.
