Conditional Inducible Protein Expression Vector (CIPE)
세포 투과성의 저분자 화합물 Shield-1은 단백질 발현의 On / Off를 제어 할 수있는 발현 벡터입니다. in vitro 및 in vivo에서
단백질의 functional analysis에 유용합니다.
CIPE-100 vector
CIPE vectors provide a cloning system where the stability of a specific protein depends on the presence or absence of a cell-
permeable biologically inert molecule Shield. FKBP-derived destabilizing domains (DDs) can typically be fused to any
protein without affecting expression and function. The fusion protein is unstable in the absence of the ligand(Shield-1), and
administration of the Shield-1 causes the fusion protein to be stabilized.
CIPE-100 vector (4620 base pairs) is available with kanamycin resistance for prokaryotic selection and neomycin resistance
for eukayonic resistance.

• 목적 단백질의 code 영역을 제한효소를 이용하여 cloning 하여, FKBP(FK506-binding protein)의 불안정화영역(Destabilized
Domain, DD) 융합 단백질을 발현합니다.
• DD 융합 단백질은 proteasome에 의해 분해됩니다. DD 융합 단백질은 Shield-1과 결합해 안정화하여, 세포 내에
축적합니다 .
• Shield-1 농도 의존적으로 단백질의 발현 수준을 제어 할 수 있습니다.
• DD 영역의 양단에 MCS (Multiple Cloning Site)가 존재하기 때문에, 목적 단백질의 N 말단 또는 C-말단 어느 것에도
DD 영역을 융합 할 수 있습니다.
• Kanamycin 와 Neomycin 의 선택이 가능합니다.
