The multipurpose antibody for analysis of GPVI
The monoclonal antibody JAQ1 reacts with the platelet collagen receptor GPVI. On the one hand, it inhibits collagen-induced aggregation of mouse platelets, whereas cross-linking of JAQ1 by a secondary antibody induces platelet activation and aggregation. JAQ1 can be used for immunoprecipitation, immunohistochemical analysis of acetone-fixed frozen sections, immunofluorescent staining, and Western blot analysis under non-reducing condtions.
JAQ1 recognizes GPVI in Western blot analysis
After separation of mouse platelet lysate by SDSPAGE (non-reducing conditions) and transfer to PVDF membrane, the membrane was incubated with JAQ1 (1-3 μg/ml) and bound antibody was visualized with HRP-conjugated anti-rat IgG and ECL.
Detection of GPVI on mouse platelets
Mouse blood was diluted 1:20 and 25 μl of this dilution were stained with 5 μl control IgG-FITC (emfret Analytics, P190-1, black line) or JAQ1-FITC (shaded area) for 15 min at RT and analyzed directly. Platelets were gated by FSC/SSC characteristics.
