1. Description :
Human PAK2 kinase (UniProt: Q13177; NCBI: NM_002577) belongs to a family of p21-activated kinases. These serine/
threonine kinases are regulated by the small GTP binding proteins Cdc42 and Rac and have been implicated in a wide
range of biological activities. PAK2 is activated by proteolytic cleavage during caspase-mediated apoptosis, and may play a
role in regulating the apoptotic events in the dying cell.
PAK2KD is a recombinant wild type human PAK2 kinase domain (amino acids 227–524 of full-length human PAK2)
expressed in E. coli and purified to near homogeneity with high specific activity.

2. Molecular Mass and Phosphorylation :
The calculated mass is 33,510. PAK2KD is fully phosphorylated at T402 and partially phosphorylated at additional sites
with measured mass of 33,593 (1P), 33,673 (2P), 33,753 (3P), and 33,834 (4P).

4. Stability and Storage :
Stable at -70℃ for 12 month from date of receipt. Protein should be thawed on ice. Protein can be flash-frozen in liquid nitrogen and stored at -70℃.

Analysis of enzymatic activity was performed according to Zlyte assay protocol (Invitrogen):
  1. Different concentration of PAK2KD were incubated in a buffer containing 50 mM HEPES (pH7.5), 10 mM MgCl2, 1 mM EGTA, 200 mM ATP, 0.01% Brij-35, and 2 µM substrate (SER/THR 14, Invitrogen) at room temperature for 1 hour.
  2. Developer solution was added to reaction and reaction was stopped after 1h of incubation at RT.
  3. Fluorescence was detected using lexc=460±40 nm and lem=528±20 nm filters.

1. Description :
TurboNuclease is a recombinant form of Serratia macescens extracellular endonuclease (encoded by the same gene of
Bezonase) produced in E. coli using a proprietary process. This nonspecific endonuclease hydrolyzes both single- and
double-stranded nucleic acids (DNA and RNA) to 5’-phosphorylated oligonucleotides of 1-4 bases in length.
TurboNuclease is a highly purified homodimer of 27 kDa subunits that has exceptional high specific activity and is free of
protease activity. TurboNuclease is ideal to digest nucleic acids and to reduce viscosity during protein purification and
sample preparation.

4. Unit Definition :
One unit of TurboNuclease converts 1.0 OD260 of salmon sperm DNA into acid-soluble nucleotides in 30 minutes at 37oC
in a reaction buffer of 50 mM Tris-HCl, pH 8.0 and 1 mM MgCl2. This corresponds to complete digestion of 50 µg of
salmon sperm DNA into oligonucleotides. TurboNuclease has similar specific activity as Benzonase

< 0.1 EU / 1,000 units of TurboNuclease as determined by the Endo-Safe LAL Assay.

50 µg of salmon sperm DNA was incubated with the indicated units of TurboNuclease and another brand of nuclease at 37oC for 30 minutes in a buffer of 50 mM Tris-HCl, pH 8.0 and 1 mM MgCl2. DNA digestion was monitored by agarose gel.
