OptiPharma™ Protein Solubility Kits help researchers to identify formulations that protect target

biopharmaceutical from aggregation, or to solubilize an aggregated biopharmaceutical sample.

 

OptiPharma Protein Solubility Screening Kit contains a combination of 10 types of buffers (pH varies from 3 to

8.5) and 13 pharmaceutical excipients (salts, amino acids, sugars, surfactants, preservatives). A total of 93

different formulations in a 96-well format can be tested in single, label-free experimental setup. The

remaining 3 wells are for control experiments. The table below lists the formulationsin the OptiPharma Kit.

Proteins aggregate, This undesired process is often caused by elevated temperature, vigorous stirring,

addition of ligands or molecular binding partners or just by mere storage of a protein sample over a period of

time. Protein aggregation can often be avoided with proper choice of pH, salt or stabilizing additives.

OptiPharma Protein Solubility Screening Kit contains a combination of 10 types of buffers (pH varies from 3 to

8.5) and 13 pharmaceutical excipients(salts, amino acids, sugars, surfactants, preservatives). A total of 93

different formulations in a 96-well format can be tested in single, label-free experimental setup. The

remaining 3 wells are for control experiments.  Soluble proteins pass filters, aggregated proteins do not. This

simple feature is exploited to in the filtration step. The wells that contain soluble proteins will yield protein

after the filtration process; those with protein aggregates will not contain any protein. After running

theOptiPharma Kit you have established conditions that keep your biopharmaceutical target in solution and

you know conditions at which your protein aggregates.

  assays, SDS-PAGE, Western blot, mass spectrometry, and fluorescence-based protein detection.

 

 

New FASOS technology

OptiPharma, similar to OptiSol, is based on proprietary FASOS Technology. FASOS stands for Filter Aided

Solubility Optimization Screening. A protein in original buffer condition is mixed separately with various

OptiPharma formulations. Protein in formulations that promote solubilization will be detected in the filtrates

after passing through a specific molecular weight cut-off filter. On the other hand, aggregated protein due to ‘

bad’ formulations is retainedon the filter. This combinatorial solution and filtration approach identifies the

optimal pH, buffer type, and additives that maintain samples solubilized. One can either solubilize an

aggregated protein solution or select a particular sample stress (heat, freeze-thaw, storage, shear etc.) and

identify those solutions that keep the protein sample solubilized without aggregation. Data evaluation is

supported with Pharma Dashboard.

 

 

 

Applications for the OptiPharma™ Protein Solubility Screening Kit

The OptiPharma™ Protein Solubility Screening Kit can be used to protect target proteins against a variety of

aggregation-inducing stresses. Proteins often aggregate when exposed to elevated temperature, when they

are stored for longer periods of time, when protein solutions are repeatedly frozen and thawed or when the

liquid is exposed to high shear forces or intense light. Oxidation by air, reactive reagents,and surface

exposure are also causes of protein aggregation. These stressors can be used to assess the susceptibility

of a particular protein sample and identify protective agents. OptiPharma™ Protein Solubility Screening Kit

can provide you answers.
