For Downstream Applications Cyanase™ can Easily be Removed From Lysate with any of the available Inactivation Resins.

Lysis Protocol:
1. Add contents of Cyanase™ nuclease tube into lysis buffer before beginning any procedures with the kit.  The
   Cyanase™ will be stable in the lysis buffer at 4°C for up to 1 year.  
2. Resuspend cell pellet up to 1 X 1011 cells in lysis buffer (up to 1 g wet cell paste per ml).  Add 1 M MnSO4 to
   resuspension for a final concentration of 6 mM.  Only add MnSO4 to lysis buffer when you are ready to begin the lysis
   as the MnSO4 is not stable long term at the lysis pH of 8.0.  
3. Pre-wash OmniLyse® cartridge in 500-100 µl of deionized water or 50 mM Tris pH 8.0.  Attach cartridge to
   Bac-Pac™ battery pack via the blue mating connectors, lining up the red arrows.  Attach the luer-lock tubing tip to
   the sample entry port and using the provided syringe, fill the chamber with buffer.  Turn on the battery pack, and
   pass the solution back and forth several times through the cartridge.  Expel all the fluid and turn off battery pack.
4. Insert the tubing tip from the sample port into the sample.  Draw Sample into OmniLyse® cartridge with attached
   syringe, displacing air.
5. Turn on battery pack and slowly draw majority of the remaining sample through the cartridge such that cartridge
   remains filled with sample.  Reverse direction, dispensing the majority of the sample through the cartridge into the
   sample tube.  Continue withdrawing and infusing the sample for 1 minute to 2 minutes, depending on the ease of
   lysis and desired lysis efficiency.  After final removal of sample from syringe, use air to purge the remaining lysis from the
   cartridge.  Turn off battery pack. Cartridges can be reused once on fresh samples within 24 hours if there are no cross
   contamination concerns.
6. Allow the lysis to incubate at room temperature for at least 5 minutes to degrade all remaining nucleic acid
   contamination.  Longer incubations can be performed if desired depending on the final level of nucleic acid removal
7. Spin down pellet and remove lysis for further processing.  
8. Alternately, Cyanase™ Inactivation resin (Cat#1010, 1020) can be added to the cell lysate after incubation and spun
   as before to remove the Cyanase from the reaction, or the cell lysis can be added to Cyanase™ Inactivation resin
   cartridges (Cat#1015) for high throughput Cyanase™ removal if nucleases are a problem for downstream applications.

Cyanase™ is fully active in DTT up to 100 mM, and most common non-ionic detergents up to 1% including Triton X-100, Tween 20, and Igepal CA630.  Cyanase™ is active in Urea up to 3 M.   Cyanase™ is unaffected by chemical lysate
treatments such as lysozyme or detergent.
Cyanase™ is inhibited by phosphate buffers at 20 mM or higher, and NaCl concentrations of 200 mM or greater.  For
optimal activity, it is recommended to use Mn2+ in place of Mg2+ and avoid any chelating reagents as the enzyme
requires divalent cations for activity.  For pH use greater than 8.3 or less than 7.0, Mg is recommended for maximum
activity due to Mn instability.  It is recommended to add Mn fresh to each reaction for maximal activity.  Do not freeze
Cyanase™ as it will result in a loss of activity of about 10-15%.  See application guide and sample protocols for usage
