ScreenFect™ siRNA is a transfection reagent composed of new cationic lipids selected by screening

experiments *1 by click chemistry. It is applicable to cell lines from various eukaryotic species and can be

directly added to medium containing serum.

Because of the low cell toxicity, medium change after transfection is not required. Furthermore, this kit does

not contain any poisonous and deleterious components.

ScreenFect™ siRNA Dilution Buffer is a Wako-recommended buffer for transfection. However, basal medium

such as D-MEM, E-MEM may be more suitable depending on cells. Please optimize dilution condition for

your cells.

 

*3:

Add ScreenFect™ siRNA Transfection Reagent into Dilution Buffer with rapid pipetting (4 - 5 times). Tap the

tube with finger for well-mixing if necessary.

 

*4:

This one-step method is applicable to not all cell types. When you does not get ideal transfection efficiency

at the protocol, remove the medium from cells, first. Then add 420 μL of new medium to SF-siRNA

complexes, mix by pipetting, and immediately apply to the cells.

NOTICE

First, dilute ScreenFect™ siRNA Transfection Reagent and siRNA with Dilution Buffer provided, respectively

  as mentioned above.

Since the siRNA transfection efficiency depends heavily on cell type, the condition, the passage number   

  and combination with siRNA transfected, please adjust the each concentration of ScreenFect™ siRNA   

  Transfection Reagent and siRNA, respectively before starting on new transfection experiment.

  ScreenFect™siRNA transfection

  condition:

  SiRNA (LPR6): 1 pmol

  Transfection Reagent: 0.1 μL

  Suspended Cells : 420 μL

 

LRP6 siRNA  was transfected into MEF. LRP expressions after 24 hours were examined by western blotting.

Knockdown effect of ScreenFect™siRNA is the same or more than competitor A and B.
